I have used Mate in the Raspberry Pi3 with alsa, I had to remove
pulseaudio because As I don't have an USB Audio Adaptor, spd didn't work
with pulse. I'm waiting for the soundcard that I've bought (it's one
from sound injector) and I'll see how it goes. I am particularly
interested in getting pulse to work because I'd like to connect the PI
to a bluetooth headset, but can't because bluez5 does not include
support for alsa.

If you want to have speech without the needing to install pispeakup, you
may find useful the images available in the downloads section of the
Raspberry Vi website:

There is raspbian jessie and it should include pispeakup, so you'll have
speech as soon as you write the image in the SD Card.
